In ATLAS Diagenode (#6) will focus on 3 different aspects of the project:
1. Optimization of random amplification of immunoprecipitated DNA.
2. Antibody production and validation.
3. Kit development and optimization for the research or diagnostic market.
Our R&D group has a strong expertise in technologies linked to Chromatin Immunoprecipitation (ChIP). We indeed produce and validate antibodies for ChIP. We also design kits for the Epigenetics research field, including ChIP kits. Our goal is to develop Epigenetics research tools. In general, ChIP is to become easier, quicker, more HTP, more efficient, more sensitive, more simple to analyse. Therefore, we are currently developing additional ChIP kits, based on the use of a lower amount of cells per ChIP reaction. Besides ChIP, we are also very much interested in other Epigenetics methods such as the study of DNA methylation and the establishment of a kit for in vitro Chromatin Assembly. All together, this demonstrates our active involvement in the development of new tools in the Epigenetics field and our great interest in ATLAS.
1- Optimization of random amplification of immunoprecipitated DNA (in WP4):
Several methods exist for random amplification of DNA. We will focus on the optimization of 2 selected methods: one that is based on PCR and another that is PCR independent. We will then compare both methods and analyze bias linked to each method. Finally we will optimize one method of amplification and the amplified products will be studied by ChIP-on-chip and/or ChIPSeq.
2- Antibody production and validation (in WP3):
We will produce antibodies directed against targets that are relevant to the ATLAS project. We will affinity-purify the antibodies and submit the antibodies to quality control (WB, ELISA, IF, ChIP).
3- Kit development and optimization for the research or diagnostic market (in WP3):
Each new ChIP procedure requires tedious optimization of several reaction conditions. Diagenode will provide optimized reagents and kit(s) adapted to the ATLAS technology. The new tools will be designed first for research use and then will be applied the more specific field of diagnosis.
