DNA extraction is one of the critical stages in the acquisition of DNA barcodes. We have developed a set of “Mix and Match” protocols allowing the extraction of DNA from various organisms including fungi and plants. These protocols are compatible with automation and deliver high quality DNA for only a fraction of the cost associated with the use of commercial kits. In addition, the voucher recovery step aids in preserving the morphology of small specimens for future reference. The same protocols can be easily down-scaled to a tube format for forensic applications and low throughput labs.
