In order to fulfill his professional commitment, Jatin (Taaha Shah) reaches the venue where he is invited to inaugurate a book. He is tensed otherwise as his elder brother Akash (Priyanshu Chatterjee) is battling for life in the ICU. No sooner he starts reading out a few pages, he finds something ambiguous about the story and immediately sets out to seek the identity of the publisher leaving the book launch midway. However to his shock, the publisher claims to be unaware about the author's whereabouts and maintains that it was couriered to her office while they decided to publish it due to its intriguing plot. The book launch triggers a series of flashback events where Jatin falls in love with Barkhaa (Sara Loren) at the very first sight and despite knowing she is a bar singer by profession, his love never fails to die. Barkhaa, who has a mysterious and troubled past, loses faith in humanity because of which she deters Jatin from marrying her. The only person whom Barkhaa trusts is her longstanding best friend Madhu (Shweta Pandit). Despite Barkhaa's warnings, Jatin goes ahead and proposes her for marriage in spite of knowing the fact that she is already a single mother of a girl child. While Jatin introduces Barkhaa to his family, his father (Puneet Issar) doesn't approve of the alliance and feels that the only person who can actually make Jatin understand the reality of life is his elder brother Akash. The mystery envelops when Jatin's family along with Akash in no time storms away this relationship and in the meanwhile Barkhaa also goes missing from the house, thus adding to Jatin's woes. Arun Mathur and his college sweetheart, Lily Fernandes, even though of different faiths, Hindu & Catholic respectively, are thrilled when their parents agree to the marriage. Ruby Fernandes welcomes Arun and encourages him to call her "mommy", while Arun's dad is already making preparations for the marriage. Then the doctor announces that Lily has blood cancer and may not have very long to live. She and her mom not only suppress this news from Arun, but also cancel the engagement ceremony. Dr. Deepak lives with his wealthy father, a Thakur, in a small town in India. While the Thakur would like to get Deepak to marry a woman from an equally wealthy family, he has fallen in love with a poor woman named Asha, and would like to marry her. His travels take him to meet a beautiful woman named Kiran, her father, and an elderly man named Doctor. Pratap is perturbed by this doctor, who seems to know him very well, and wants him to leave his job at the chemical company, and start practicing medicine in the small town where they live. What Pratap does not know that this doctor is none other than his biological father, Deepak. What both don't know is what steps the Thakur will take when he finds out that Pratap is none other than his grandson.
Yuvraj Vijay Singh (Salman Khan) a prince of Pritampur, is soon to be crowned as king and is engaged to Princess Maithili (Sonam Kapoor), but due to his stiff and stubborn nature Vijay faces many problems with Maithili as well as with his brother and sisters. Yuvraj Vijay barely escapes a plan coordinated by Yuvraj Ajay and Chirag to kill him but gets badly injured. He's hidden in a secret chamber maintained by two doctors.
Meanwhile, Vijay's lookalike Prem Dilwale (Salman Khan) a perky, lively and carefree stage actor who falls in love with Princess Maithili, reaches Pritampur to meet her along with his friend Kanhaiya (Deepak Dobriyal). At a bus stop, the Security Head of Pritampur Palace, Sanjay (Deepraj Rana) notices him by chance, and takes him to Diwan Sahib (Anupam Kher). Upon witnessing the striking resemblance between Prem and Yuvraj Vijay, diwan asks Prem to take Yuvraj Vijay's place while Yuvraj recovers from coma
Chirag tries to shoot them down but falls to his death. Ajay regrets his deeds, while Vijay reconciles with his family. Maithili is shocked to know the truth about Prem and realizes that she loves Prem not Vijay, while Prem returns to his home. At the end, the royal family reaches Prem's house to reunite Prem and Maithili.

Rajesh (Mohnish Bahl) runs the business for his dad, Kailashnath (Alok Nath), and is an eligible bachelor. The family wants him to marry Pooja Choudhury (Renuka Shahane), to which he agrees.
